The Challenge: Hidden Inefficiencies in Pharmaceutical Compression In a high-stakes manufacturing environment, even minor waste can lead to major financial leaks. At our Brookhaven facility, we identified a critical pain point in the compression process: a consistent yield loss of approximately 5 kg per batch. This wasn’t just a waste of material; it resulted in 41 deviations over a two-year period, leading to back-orders and a high risk of “Failure to Supply” (FTS). At the time, the process was operating at a Sigma Level II.

The Strategy: Data-Driven Operational Excellence Using a Six Sigma methodology, our team conducted a deep dive into the root causes of yield loss. Through Gemba walks and Pareto analysis, we discovered that the “standard” practice for QA sampling was actually a primary driver of waste.

We proposed a Compliance-Driven Reduction in Bulk Retain Sampling. By analyzing FDA Guidance (21 CFR 211.170(b)), we confirmed that the current practice of collecting 3x the QC sample size was not a regulatory requirement for intermediate stages.

The Solution: Lean Sampling Protocols

  • Regulatory Alignment: Transitioned from a 3x sample size to a 1x QC sample size for in-process bulk retains.
  • Operational Optimization: Revised QA procedures to save approximately 650 tablets per batch.
  • Compliance Integrity: Ensured all necessary data for investigations remained available through Finished Product (FP) retain samples, which are the FDA-required standard.

The Bottom Line Results The impact of these lean improvements was immediate and measurable:

  • Hard Manufacturing Savings: A minimum of $302,304 saved in 2024 at a single facility.
  • Top-Line Impact: The sales value of the recovered yield reached $952,833.
  • Future Projections: With increased production and price adjustments, we are on track for $1.14M in savings for 2025.
  • Supply Chain Resilience: Significant reduction in potential “Failure to Supply” incidents by avoiding unnecessary deviations and back-orders.
